The Dynamic Role of Human Primary Endothelial Cells in Human Physiology

Human primary endothelial cells have been employed extensively in pathophysiology and therapeutic research. These cells are the basic unit of any blood vessel—capillaries, arteries, and veins. As a core component of the circulatory system, they are integrated into each organ. Their role extends from being a mere barrier to regulating tissue homeostasis, vessel integrity, disease pathogenesis, immune modulation, etc. The significance of endothelial cells in tissues has even led to their incorporation into 3D structures.

Human liver hepatocytes are an in vitro tool for studying liver physiology. Due to their retention of tissue properties, especially drug detoxification, their use has notably increased in pharmacological and toxicological studies. Evaluating drug toxicity and the resultant metabolite on primary human hepatocytes has facilitated effortless and high-throughput screening of multiple drug candidates simultaneously. They are considered the gold standard for preclinical drug validation assays. They have also augmented our understanding of liver disorders. However, they have limited proliferative capacity in vitro. Their applications have surged their in vitro use, prompting scientists to develop strategies for better hepatocyte culture.

Mouse kidney epithelial cells are primary cells derived from mouse kidney tissue, commonly used in research to study kidney function, drug transport, and toxicology. These cells closely mimic natural kidney physiology but have a limited lifespan in culture, which can restrict long-term experiments.

Immortalized mouse kidney epithelial cells, on the other hand, are genetically modified to proliferate indefinitely while retaining only few characteristics of normal kidney cells. This makes them valuable for consistent, reproducible studies over extended periods. They are widely used in nephrology research, including studies on kidney disease mechanisms, drug screening, and epithelial cell behavior under various experimental conditions.

Endothelial cells line the blood vessels and, therefore, are vital for hemostasis. They are also essential for the transport of cells across the vessels and for maintaining a barrier for the underlying tissue. Their role in inflammation, adhesion, and coagulation has gained attention due to their implication in vascular disorders. Their ubiquitous presence can turn a localized disease into a systemic one. Research on human primary endothelial cells has revealed the adaptation of these cells to tissue. Therefore, each tissue-specific endothelial primary cell is unique in its phenotype and genotype. The varied surface markers can be used to identify subpopulations and target therapeutics. Previously considered as a static barrier, endothelial cells have emerged as key players in health and disease. The scientific studies have grown substantially to develop novel therapeutics based on these cells.
